Hydroxyproline and Collagen Analysis in Meat Products

Quality assessment in meat and meat products is not limited to total protein content. In particular, the amount of connective tissue-derived proteins is important for evaluating product composition and compliance with legislation. One of the main analytical indicators used for this purpose is hydroxyproline content.

What Is Hydroxyproline?

Hydroxyproline is a characteristic amino acid found in high amounts in the structure of collagen. It plays an important role in stabilizing the three-dimensional structure of collagen and is therefore used as an analytical indicator for determining the amount of connective tissue in meat products.

In the analysis, the hydroxyproline content is measured directly, and the collagen content is calculated based on the result obtained:

Collagen = Hydroxyproline × 8

This provides quantitative information about the amount of connective tissue-derived protein in the product.

Why Is Collagen Important in Meat Products?

A high protein content in meat products does not always mean that all of the protein originates from muscle tissue. Since collagen is found predominantly in connective tissue, the proportion of collagen within total protein helps assess the balance between muscle meat and connective tissue in the product.

The main reason legislation limits the collagen connective tissue protein ratio is to prevent excessive use of connective tissue, maintain product quality, and ensure that the meat product offered to consumers has the expected composition.

Why Is It Important Under the Meat Communiqué?

Under the Turkish Food Codex Communiqué on Meat, Prepared Meat Mixtures and Meat Products (2026/15), published on 16 September 2026, the collagen connective tissue protein / meat protein ratio is defined as a direct compositional criterion for certain product groups.

For example, the maximum limits are:

  • 10% for prepared poultry meatballs and burgers,
  • 20% for sucuk,
  • 25% for heat-treated sucuk,
  • 25% for emulsified meat products.

In addition, since minced meat labels are required to include the statement “Collagen connective tissue protein / meat protein ratio: max. …”, this parameter has also become important for product specifications and labelling processes.

How Is Hydroxyproline Analysis Performed?

At Bilim Laboratories, hydroxyproline determination in meat and meat products is performed within the scope of TS 6236 ISO 3496.

During the analysis, the sample is hydrolysed, the released hydroxyproline is converted into a coloured compound through defined chemical reactions, and its quantitative determination is carried out spectrophotometrically. Based on the result obtained, the collagen content and, where required, the collagen connective tissue protein / meat protein ratio can be calculated.
